Abstract:
My group works on epigenetics and non-coding RNAs using a combination of biochemical and genetics approaches including cell culture and animal model systems and genome-wide approaches. In this seminar I will give a brief overview of our work and focus in on a microRNA project in which we demonstrate important roles for miR-9 in Hodgkin's Lymphoma. Briefly, we find miR-9 to regulate cytokine signaling via the RNA binding protein HuR and to regulate splicing via direct targeting of the splicing factor SFRS1 and the nuclear non-coding RNA MALAT1. Interestingly, splice junctions affected by miR-9 are predominantly found in ribosomal proteins and translation factors and resultantly miR-9 affects the rate of translation. Importantly, systemic inhibition of miR-9 in a xenograft model system results in reduced tumor outgrowth pointing to a causal role for miR-9 in tumorigenesis and a therapeutic potential of inhibiting miR-9.
