Comment	Four cysteine desulfurases have been identified in B. subtilis: NifS, YrvO, IscS, and CSD. NifS has been associated with nicotinamide biosynthesis and is an essential gene \|CITS: [8444804]\|. CSD may be involved in iron sulfur cluster assembly, as it is adjacent to yurV, an IscU homologue, YrvO is localized adjacent to tRNA (5-methylamino)methyl-2-thiouridylate methyltransferase, and iscS is adjacent to \|FRAME: BSU29580\|, suggesting a link to thiamin biosynthesis. All four cysteine desulfurases were analyzed for their efficiency in catalyzing the transfer of sulfur from cysteine to ThiS, and while all were able to catalyze the reaction, IscS was the most efficient, followed closely by NifS, YrvO, and CSD \|CITS: [14567704]\|.

	(Etha) Cells were grown in a synthetic medium (J. Stülke, R. Hanschke, M. Hecker, J Gen Microbiol 139, 2041, Sep, 1993) with 0.2 % glucose as carbon source (Belitsky Minimal Medium/BMM) at 37 °C with vigorous shaking. Where necessary, carbon source solutions were pH neutralized with 4 M NaOH prior to addition to the medium. Cells were harvested during the exponential growth phase.

	(S2) Cells were grown in CH medium at 37°C and sporulation was induced by resuspension in warm sporulation medium as described by Sterlini and Mandelstam (J. M. Sterlini, J. Mandelstam, Biochem J 113, 29, Jun, 1969). The initiation of sporulation was designated T0, the time of resuspension.
